The recent development of liquid biopsies has opened up new directions for monitoring the progression of cancer and the formation of metastasis, bringing us one step closer to effective personalised therapies. Despite these remarkable breakthroughs, metastasis remains the leading cause of cancer-related deaths. Therefore, it is of great significance to investigate the biology of metastasis and identify the critical steps and factors that need to be targeted in order to successfully develop efficient anti-metastatic treatments. Our work broadly aims to understand how metastasis is regulated by the circadian rhythm. While several studies have shown that the disruption of the circadian rhythm and the resulting misalignment of sleep-wake cycles promote tumour growth, our recent ground-breaking discovery indicates that the metastatic spread of breast cancer occurs during sleep. To further explore how the circadian rhythm drives metastasis, we focus on in vivo mouse models and employ cutting edge microfluidics and robotic technologies, genetic engineering, next generation sequencing and in vivo imaging systems.

• Use of fly and mammalian models to explore therapeutic mechanisms & develop new compounds for clinical trials to improve treatment for cancer patients.

• Generating highly complex Drosophila models to explore the biology of cancer therapeutics, including for thyroid, colorectal, lung, salivary gland, and breast cancers, used as the start of an integrated approach towards therapeutics.

• These models are designed to capture the genomic complexity of individual human colorectal, thyroid, lung, and breast cancer patients.

• Exploring the relationship between genomic complexity and drug resistance.

• Use of a novel platform—combining fly genetics with medicinal and computational chemistry—to develop lead compounds that attack the cancer network in the context of the whole animal. These ‘Tumor Calibrated Inhibitors’ represent a new way towards addressing drug liabilities including resistance and toxicity.

• How metabolic pathways and certain signalling nodes such as the RUNX/CBFβ transcriptional complex and pro-survival factor MCL-1, contribute to tumour progression and metastasis.

• It has been shown that high expression of RUNX1 and RUNX2 in breast cancer correlates with specific subtypes of the disease and with poorer patient prognosis. We are now investigating the functionality of these genes in epithelial cancers and have shown that RUNX2 has a role in mammary stem/progenitor cells.

• Specific interest in translational research, led by key Clinical Academics, seeking direct impact on treatment of breast cancer patients, and also aiming to advance more personalised approaches to the treatment of early breast cancer

• Research to further understand the dichotomous roles of immune cells and how tumours control immune cell behaviour. Studied in the context of breast, pancreatic and colorectal cancers. Particular interested in γδ (‘gamma delta’) T cells, a rare population of T cells with properties that are distinct from conventional CD4 and CD8 T cells, as γδ T cells can be both pro-tumourigenic and anti-tumourigenic. Ultimate goal is to understand how γδ T cells and other immune cells participate in the metastatic process and to develop new immunotherapies that counteract metastatic lesions.

• Integrins are cell adhesion receptors involved in mediating the invasive migration, growth and metastasis of cancer cells. Data from our lab and our collaborators has established that Rab-regulated integrin trafficking contributes to cancer cell migration. Have obtained detailed descriptions of the molecular machinery of integrin transport and determined how this contributes to invasive migration of cancer cells through three-dimensional matrices.
