• Focus on the dichotomous roles of immune cells and how tumours control immune cell behaviour. Particularly interested in γδ T cells, a rare population of T cells with properties that are distinct from conventional CD4 and CD8 T cells, as γδ T cells can be both pro-tumourigenic and anti-tumourigenic. Ultimate goal is to understand how γδ T cells and other immune cells participate in the metastatic process and to develop new immunotherapies that counteract metastatic lesions.
• Understanding how gene expression is regulated in health & disease, to use the knowledge to develop and refine therapeutic approaches. Focus on RNA cap, a structure found on pre-mRNA and non-coding RNA that protects RNA and recruits factors involved in processing and translation. A series of RNA capping enzymes catalyses cap formation, predominantly as the transcript is being synthesised. Interest in how these RNA capping enzymes are regulated during development and in the adult, and the impact that this has on gene expression. Investigating RNA cap regulation in embryonic stem cell differentiation, during T cell activation and following oncogene dysregulation in cancer models.
• The Translational Pharmacology & Immunology (TPI) Lab in SCS is at the heart of translational research and clinical biomarker discovery. TPI aims to foster biomarker discovery by using precision immunology. Specifically, TPI unleashes a combination of cutting - edge proteomic approaches (e.g. spatial immune profile, spectral cytometry and ultrasensitive analytical measurement of chemokines and cytokines), performed according to GCLP standards to: (i) discover patterns of immune evasion responsible for resistance to immunotherapy and additional therapeutic interventions; (ii) foster the development of novel immunotherapeutic treatments.
• Focus on co-evolutionary mechanisms in the tumour microenvironment. Cancers to do evolve in isolation. It emerges and thrives in the context of its microenvironment, engaging in dynamic cross-regulatory interactions with surrounding cells. There are laboratories in SCS that centre on uncovering the co-evolutionary mechanisms that shape tumour initiation, tissue remodelling, and progression, including the investigation of immune dysregulation. Chronic inflammation disrupts immune homeostasis, leading to immune suppression and the recruitment of tumour-promoting immune cells, such as pro-remodelling macrophages and regulatory T cells (Tregs). By studying how tumours exploit immune-stromal interactions, we aim to identify new therapeutic strategies targeting the tumour ecosystem to restore immune balance
