Professor Nicol Keith

Research interests

Professor Nicol Keith is group leader for the Telomerase Therapeutics Group. Their principal aim is to acquire an understanding of the mechanisms underlying cancer cell self-renewal. The program brings together key strategic areas of cellular immortality, cancer stem cell biology, and chromatin regulation of gene expression. Ultimately the objective is to provide rational scientific foundations for application of small molecule and gene based therapies to the treatment of human cancer.

A suitable target for development of novel anti-cancer strategies is telomerase, a ribonucleoprotein reverse transcriptase that extends human telomeres by a terminal transferase activity. Basic telomerase activity is dependent on regulated expression of the essential hTR and hTERT genes. Although encouraging progress has been made in evaluating telomerase in diagnostic and prognostic settings, perhaps the most promising potential clinical approach lies in the field of novel therapeutic strategies. The unique biology and function of telomerase, together with its complex regulation, means that therapeutic targeting is possible at various points of the cell signalling and transcriptional machinery. Thus, telomerase is not one target but many. There are different therapeutic approaches for each target, and each has its own specific advantages and limitations.

The group’s research can be visualised as two inter-related activities centred on investigating mechanisms of gene regulation and the application of this information to gene therapy strategies.
