My research focuses on companion animal virology and immunology with a view to understanding disease pathogenesis, correlates of immunity to infection and the development of novel approaches to the diagnosis, prevention and treatment of viral diseases. This work has both comparative and veterinary significance.
 
A longterm interest has been the study of feline immunodeficiency virus (FIV), an important feline pathogen bearing striking similarities to the human lentivirus (HIV), the causative agent of AIDS.  A veterinary vaccine for FIV is available in several countries outside Europe and we believe that by dissecting the protective mechanisms induced by FIV vaccines, as well as the deleterious responses induced by vaccines that enhance infection, we can advise the design of improved, novel vaccines for both human and veterinary lentiviral infections.
 
For veterinarians with feline patients infected with FIV, many questions remain as to why some cats develop AIDS rapidly while disease progression is slow in other cats. By participating in the European Advisory Board on Cat Diseases (ABCD), we contributed to guidelines, which translate the current best evidence into recommendations and advice for veterinarians dealing with feline infectious diseases in clinical practice (www.abcd-vets.org). ABCD recommends that cats should not necessarily be euthanised following the diagnosis of FIV infection as many infected cats respond well to prompt, intensive treatment for clinical signs arising indirectly from FIV infection.
 
The discovery on host-encoded mediators of intrinsic immunity to viral infection has opened up a new avenue for investigation. Factors such as APOBECs, TRIM5α and tetherin restrict lentiviral replication and areup-regulated in the host by interferons produced during the innate immune response, creating an antiviral state in previously susceptible cells. The activity of such restriction factors delays viral replication long enough to allow the host to mount an immune response.  Our hope is to define the host and virus-specific factors contributing to the pathogenesis of FIV infection and to compare the relative contributions of the intrinsic, innate and adaptive arms of the immune response towards the control of viral replication.
