Our lab is involved in two major lines of research:

Molecular Basis of Virulence in Staphylococcus aureus
Bacteria are successful as commensal organisms or pathogens in part because they adapt rapidly to selective pressures imparted by the human and animal hosts. Mobile genetic elements (MGEs) play a central role in this adaptation process and are a means to transfer genetic information (DNA) among and within bacterial species. Importantly, MGEs encode putative virulence factors and molecules that confer resistance to antibiotics. Inasmuch as bacterial infections are a significant problem worldwide and continue to emerge in epidemic waves, there has been significant effort to understand the agents that effect DNA movement.
Staphylococcus aureus, a major nosocomial pathogen, causes a wide variety of infections, from simple abscesses to fatal sepsis, plus toxinoses, such as food poisoning and toxic shock syndrome. These actions depend upon the production and secretion of over thirty specific pathogenicity factors, including superantigen toxins, hemolytic cytotoxins, tissue-component-degrading enzymes and surface proteins, that interfere with host defences. S. aureus pathogenic versatility is also compounded by its ability to develop resistance to new antibiotics almost as fast as they are introduced.
Genes for staphylococcal virulence factors are often borne by MGEs, including plasmids, prophages or pathogenicity islands. In recent years, we have extensively characterised a family of pathogenicity islands in Staphylococcus aureus, SaPIs, which contribute substantively to horizontal gene transfer, host adaptation and virulence. SaPIs represent the first pathogenicity island for which mobility has been shown. During the last few years, we have deciphered the fascinating mechanisms involved in the transfer of these mobile genetic elements. SaPIs are 15 kb or more in size, are excised, caused to replicate, and then encapsidated with high efficiency after induction by certain staphylococcal phages. This results in an extremely high frequency of transduction. Recently, we have found that similar structures are also present in pathologically relevant Gram-positive bacteria, thus defining a new family of mobile genetic elements, that we have named PICIs (for phage-induced chromosomal islands). The broad distribution of these elements suggests a relevant role in virulence. We are now analysing the functions of these elements, the extent of their dissemination and the spectrum of pathogenicity genes carried by them. Since the cell cycle of these elements require induction by bacteriophages, which in addition also have a relevant role in pathogenesis, we are also focussing in the characterization of these unexplored genetic elements.

dUTPase Signalling: from Phage to Eukaryotes
Preserving DNA integrity is of vital importance for all organisms. Genomes of all free-living organisms and many viruses encode the enzyme dUTPase (dUTP pyrophosphatase; DUT; EC 3.6.1.23), which cleaves dUTP into dUMP and pyrophosphate. Since most DNA polymerases cannot distinguish between thymine and uracil, it has been traditionally assumed that DUTs are essential for DNA integrity and viability in many organisms by reducing the dUTP pool, preventing incorporation of dUTP into the chromosome. Recently, however, DUTs have been identified as being involved in the control of relevant cellular processes. How these regulatory functions are controlled remains unsolved. Based on our recent data, our group pursues the hypothesis that DUTs are signalling molecules, from phage to eukaryotes, acting analogously to eukaryotic G-proteins. The proposed work is important as it identifies a putative new universal family of signalling proteins, which remarkably involves dUTP as a second messenger.
