The ‘antiviral state’ induced by interferons attenuates the replication of most, if not all, mammalian viruses. Upon sensing a pathogen, interferon signaling results in the upregulation of hundreds of different genes. These interferon stimulated genes (ISGs) are able to mediate further signaling, modulate the cellular environment, or interfere with viruses directly, in order to impede viral replication. Despite intensive investigation, the exact contribution of individual ISGs to the antiviral state is often unknown. However, in some remarkable cases individual ISGs have been described that effectively render cells resistant to certain viruses. The extraordinary ability of these factors to restrict virus replication can protect individuals, populations and entire species from specific pathogens.

It is my belief that many restriction factors remain unidentified because evolution has rendered viruses supremely adept at avoiding and disabling the arsenal of intrinsic immune effectors within the host cell. My lab is focused on identifying and characterising these novel intrinsic immune factors. We mainly investigate a diverse range of retroviruses where multiple antiretroviral ISGs, such as APOBEC3G, TRIM5, tetherin and SAMHD1 have redefined our understanding of species-specific restriction. Based on this experience we utilize both human and rhesus macaque ISG-expression libraries to investigate retroviral replication. We are also diversifying our approach and applying medium- throughput ISG expression screening to an expanding array of virus families.

Our long-term goal is to harness the information gleaned from characterising host-viral interactions to identify novel therapeutic targets and develop biotechnologies, based upon intrinsic immune factors, that could reduce the disease burden in human or livestock populations.
